Synthesis and Process Research of β-Asaronol by Catalytic Hydrogenation Combined with SFC

Abstract:The crude (Z)-3-(2,4,5-trimethoxyphenyl)prop-2-en-1-ol (β-asaronol) was synthesized from 3-(2,4,5-trimethoxyphenyl)prop-2-yn-1-ol by optimizing Lindlar catalytic hydrogenation, under the conditions of mass ratio of catalyst to reactants 0.2, methanol as solvent, reaction time 4 h and room temperature. Then high purity β-asaronol was obtained by SFC purification. The conversion of raw materials was more than 98%. After purified by supercritical fluid chromatography (SFC) method, β-asaronol was obtained in 74%-81% total yield with good purity (>99%). The synthesis and purification process is stable and reliable, which provides material support for further study on the biological activity of β-asaronol.
