Abstract Drug-loaded hydrogel dressings can prevent bacterial infection and eliminate inflammation with the controlled drug release, which are beneficial for the wound healing. The properties of drug-loaded hydrogels as wound dressings are determined mainly by the materials and preparation method of the hydrogels, as well as the drug loading process. In this work, 3-aminophenylboronic acid-grafted sodium alginate (Alg-PBA) was cross-linked with polyvinyl alcohol (PVA) to prepare hydrogels. The formation mechanism of hydrogels was discussed, and the effects of component ratio and PVA molecular weight on the morphologies and rheological behaviors of the hydrogels were investigated. Doxycycline hydrochloride (Doxy) was loaded onto hydrogels using supercritical solution impregnation (SSI) technology. Under the condition that the mass ratio of Alg-PBA to PVA was 2:1, hydrogels with good mechanical properties were obtained by cross-linking Alg-PBA and PVA with molecular weight of 77 kDa. The in vitro drug release study indicated that Doxy was controllably released from the hydrogel under a reactive oxygen species (ROS) responsive manner. This study provides a reference for the preparation of hydrogel wound dressings with complex structures and diverse properties.
