Synthesis Process of 2-(3-(4-(tert-butyl)phenyl)-1H-1,2,4-triazol-5-yl) Acetic Acid

Abstract:2-[3-(4-tert-butylphenyl)-1H-1,2,4-triazol-5-yl]acetic acid was synthesized using 4-tert-butylbenzohydrazide via hydrazinolysis, condensation and the cyclization-hydrolysis reaction. In hydrazinolysis reaction, toluene is used as solvent to azeotropically distill off methanol and water which are the by-products, so as to promote the reaction forward and improve the speed. In condensation reaction, the product is insoluble in the solvent ethyl acetate, and thus will precipitate from the system at the reaction temperature, strengthening the process of reaction-separation. The proposed synthesis route is simple with mild reaction conditions and the total yield of the final product is 87.8% under optimized conditions.
