Abstract  Organic room temperature phosphorescent(RTP)materials have triplet excitons with a long luminescence lifetime, with the advantages of low toxicity, good biocompatibility, and good designability compared to metal phosphorescent systems, holding unique potential in bioimaging applications. In recent years, a series of organic RTP materials with high luminance and long lifetimes have been obtained through phosphorescent molecular-structure design and aggregation-behavior modulation, laying a foundation for preparing biocompatible and stable organic RTP nanomaterials. In this review, the construction strategies and mechanism of amphiphilic molecular wrapping, host-guest interaction, nanopolymerization, and organic-inorganic nanohybridization of RTP nanomaterials were systematically summarized, the focus was on summarizing the developed RTP-stabilized nanoaggregate systems and their special photophysical properties and showcasing the unique biological applications of phosphorescent nanomaterials in fields such as cell confocal imaging, tissue hypoxia imaging, lifetime imaging and in vivo long-afterglow imaging. The prospective development direction of organic room temperature organic RTP nanobiomaterials was also discussed.
